NORTH CHICAGO, Ill. and MADISON, Wis., Dec. 13, 2024 /PRNewswire/ -- AbbVie (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/ABBV#NYSE">ABBV</a>) and Nimble Therapeutics today announced a definitive agreement under which AbbVie will acquire Nimble, including its lead asset, an investigational oral peptide IL23R inhibitor in preclinical development for the treatment of psoriasis and a pipeline of other novel oral peptide candidates with potential across several autoimmune diseases. Additionally, AbbVie will acquire Nimble's peptide synthesis, screening, and optimization platform, which uses proprietary technology to help drive rapid discovery and optimization of peptide candidates for a range of targets.
Nimble's preclinical-stage IL23R inhibitor is an investigational oral therapy for the treatment of psoriasis and inflammatory bowel disease (IBD). IL23R is a clinically validated therapeutic target in certain autoimmune diseases and a major contributing factor to psoriasis and IBD pathogenesis and progression through increased inflammation and amplified immune responses.
Under the terms of the agreement, AbbVie will make a cash payment of $200 million at closing to acquire Nimble, subject to certain customary adjustments, in addition to certain interim funding payments. Nimble's shareholders remain eligible for a potential payment, subject to the achievement of a development milestone. The transaction is subject to the satisfaction of customary closing conditions, including the applicable waiting period under the Hart-Scott-Rodino Antitrust Improvements Act.

NORTH CHICAGO, Ill., Dec. 9, 2024 /PRNewswire/ -- AbbVie (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/ABBV#NYSE">ABBV</a>) today announced new results from two ongoing clinical trials evaluating epcoritamab, a CD3xCD20 bispecific T-cell-engaging antibody administered subcutaneously, in adult patients with diffuse large B-cell lymphoma (DLBCL) at the 66th Annual Meeting and Exposition of the American Society of Hematology (ASH).
Arm 1 of the Phase 1b/2&nbsp;EPCORE&#xAE; NHL-2 multi-arm trial evaluates fixed-duration investigational epcoritamab in combination with rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone (R-CHOP) in untreated high-risk DLBCL patients (n=46) with International Prognostic Index (IPI) scores of 3 to 5 (Abstract #581).1&nbsp; Results from this arm of the study showed an overall response rate (ORR) of 100% and a complete response (CR) rate of 87%. Among complete responders, an estimated 83% remained in remission after two years. Separately, three-year follow-up results from the Phase 2 EPCORE&#xAE; NHL-1 trial (Abstract #4480),2&nbsp; evaluating epcoritamab monotherapy in challenging-to-treat adult patients (n=157) with relapsed or refractory (R/R) large B-cell lymphoma (LBCL) after two or more lines of systemic therapy showed that among the 41% of patients who achieved a CR, an estimated 52% were still responding at three years (median CR duration: 36.1 months).
DLBCL is the most common type of non-Hodgkin's lymphoma (NHL) worldwide, accounting for approximately 25-30% of all NHL cases.3,4 In the U.S., there are approximately 25,000 new cases of DLBCL diagnosed each year.5 DLBCL can arise in lymph nodes as well as in organs outside of the lymphatic system, occurs more commonly in the elderly and is slightly more prevalent in men.6,7 DLBCL is a fast-growing type of NHL, a cancer that develops in the lymphatic system and affects B-cell lymphocytes, a type of white blood cell. For many people living with DLBCL, their cancer either relapses, which means it may return after treatment, or becomes refractory, meaning it does not respond to treatment. Although new therapies have become available, treatment management can remain a challenge.6,8

The trial met its primary endpoint – patients treated with tavapadon experienced a statistically significant reduction (improvement) from baseline compared to placebo (placebo: -1.2; tavapadon 5-15 mg: -10.3; p-value &lt;0.0001 versus placebo) in the Movement Disorder Society &#8211; Unified Parkinson's Disease Rating Scale (MDS-UPDRS) Parts II and III combined score at week 26.
The MDS-UPDRS was developed to evaluate various aspects of Parkinson's disease including non-motor and motor experiences of daily living and motor complications.
The TEMPO-2 trial also met the key secondary endpoint, demonstrating a statistically significant and clinically meaningful improvement in motor aspects of experiences of daily living (MDS-UPDRS Part II) in the tavapadon group (5-15 mg) compared to placebo at week 26.
The safety profile observed in the TEMPO-2 trial was consistent with prior clinical trials. The majority of adverse events reported were mild to moderate in severity.
Full results from the TEMPO-2 trial will be submitted for presentation at a future medical meeting. AbbVie is on track to submit the New Drug Application to the FDA in 2025.

NORTH CHICAGO, Ill., Dec. 9, 2024 /PRNewswire/ -- AbbVie (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/ABBV#NYSE">ABBV</a>) today announced positive topline results from its pivotal Phase 3 TEMPO-2 trial evaluating investigational tavapadon as a flexible-dose monotherapy in early Parkinson's disease. Tavapadon is the first and only D1/D5 partial agonist under investigation as a once-daily treatment for Parkinson's disease.
The TEMPO-2 trial evaluated the efficacy, safety and tolerability of a flexible-dose (5 mg to 15 mg, once daily) treatment with tavapadon as a monotherapy in adults with early Parkinson's disease. The trial met its primary endpoint – patients treated with tavapadon experienced a statistically significant reduction (improvement) from baseline compared to placebo (placebo: -1.2; tavapadon 5-15 mg: -10.3; p-value &lt;0.0001 versus placebo) in the Movement Disorder Society - Unified Parkinson's Disease Rating Scale (MDS-UPDRS) Parts II and III combined score at week 26.1
The TEMPO-2 trial also met the key secondary endpoint, demonstrating a statistically significant and clinically meaningful improvement in motor aspects of experiences of daily living (MDS-UPDRS Part II) in the tavapadon group (5-15 mg) compared to placebo at week 26.1

On Monday, Alector, Inc.&nbsp;(NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/ALEC#NASDAQ">ALEC</a>) released results from the INVOKE-2 Phase 2 trial evaluating the safety and efficacy of AL002 in slowing disease progression in individuals with <a target="_blank" href="https://www.benzinga.com/pressreleases/24/11/g42174676/alector-announces-results-from-al002-invoke-2-phase-2-trial-in-individuals-with-early-alzheimers-d" rel="noreferrer noopener">early Alzheimer&#8217;s disease (AD)</a>.
AL002 failed to meet the primary endpoint of slowing Alzheimer&#8217;s clinical progression as measured by the Clinical Dementia Rating Sum of Boxes (CDR-SB).
Also, no treatment effects favored AL002 on secondary clinical and functional endpoints.
The company says treatment with AL002 resulted in sustained target engagement and pharmacodynamic responses indicative of microglial activation.
Similarly, there were no significant effects on Alzheimer&#8217;s fluid biomarkers favoring AL002, and amyloid PET imaging demonstrated no treatment-related reduction of brain amyloid levels.
As previously reported, INVOKE-2 showed MRI changes resembling amyloid-related imaging abnormalities (ARIA) and infusion-related reactions. ARIA was primarily seen in participants treated with AL002.
Based on the results, Alector is stopping the long-term extension study.
Alector&#8217;s focus are progranulin-elevating programs, latozinemab, and AL101/GSK4527226, developed in collaboration with GSK Plc (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/GSK#NYSE">GSK</a>).
Topline data from the pivotal INFRONT-3 Phase 3 trial of latozinemab in frontotemporal dementia with a progranulin gene mutation is expected in late 2025 or early 2026.
PROGRESS-AD Phase 2 trial of AL101/GSK4527226 in early AD reached over one-third of its target enrollment of 282 participants. Alector is also advancing its preclinical candidates aimed at a broad and diverse range of protein and enzyme targets.
To align resources with these strategic priorities, Alector is reducing its workforce by approximately 17%.
As of September 30, 2024, Alector has $457.2 million in cash, cash equivalents, and investments, which the company continues to expect will provide a runway through 2026.
William Blair expressed surprise and disappointment at the update, given the strong scientific basis, evidence of target engagement, and potential signs of disease modification tied to ARIA cases. The analyst maintained the <a target="_blank" href="https://www.benzinga.com/quote/ALEC/analyst-ratings" rel="noreferrer noopener">Outperform rating.</a>
Despite still seeing promise in TREM2 as a target and in neuroinflammatory modulation, the firm is removing AL002 from its model due to the study&#8217;s failure, underscoring the difficulties in developing treatments for neurodegenerative diseases.
The analyst also expects AbbVie Inc (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/ABBV#NYSE">ABBV</a>) to opt out of the program partnership and forgo the potential <a target="_blank" href="https://www.benzinga.com/pressreleases/24/11/g41786512/alector-reports-third-quarter-2024-financial-results-and-provides-business-update" rel="noreferrer noopener">$250 million licensing payment</a>.
